We are comparing the germination rate of sterilized (70% EtOH with 1% Tween-20) and unsterilized seeds. The vernalized seeds were placed under LED light for germination and covered with transparent plastic sheets to keep them moist.

  1. Germination rate of Soybeans and Broad beans were bad in sterilized seeds (about 10% for both) and non sterilized seeds (only soybean about 20%). According to literature, soaking the seed in water for 8 to 12 hours improve germination rate of soybean. I will try to sterilize the seeds and then soak the seeds in water for over night (or better be other way around??). The idea behind is that when you sterilize the seeds, they soak up the sterilization buffer which might be influencing the germination rate.
